Myers/Pyramid and I think Sadako if you count the novels and stuff.
Weakest is probably Legion and Ghostface. They're just some kids and a dude with a knife.

Probably Blight and Vigo.
Blight originally wasn't a Killer or beholden to any of the Entity's rules, he basically had free reign of the realm - he would go through trials and through other places we hsven't seen in-game that are outside the 'trial areas' we see when we play. But he was still trapped in the realm, so he tried to figure a way out for himself - resulting in all his twisted experiments upon himself and the other Killers.
However, things didn't pan out for him, because he grew addicted to the Blight substance and eventually turned into a semi-human monster that the Entity could easily control via it's overpowering addiction to Blight.
Vigo was an older lore character and the creator of a lot of Survivor Offerings as well as the Hatch. While not playable, they are a Survivor and were insightful/powerful enough to be able to manipulate the entire format of the Entity's trial rules.
Like Blight, Vigo also discovered the Blight substance and experimented on it. (In fact, Blight even used their notes in his research). In the early days of the lore, it was thought maybe they actually escaped, but IIRC the decs confirmed they didn't in a lore stream way, way back. We don't much else about them and AFAIK the lore has largely abandoned them to focus more on playable characters and the Observer.
I could be wrong because I haven't read into the lore a ton lately, but those two characters have had the most agency in the Entity's Realm, despite being confined.
As for weakest, there's not much distinction. All of the Survivors are more or less trapped in an endless nightmare they can't wake up from. All of the Killers have either been manipulated or tortured by the Entity, or they serve it or made a pact with it.
In the end, they're all just prisoners with no hope of escape. Some of them just have different perspectives on their prison.

Strongest survivor would be Chris Redfield for me, because nobody else has been recorded to punch a boulder to move it.
Strongest killer would be Pyramid Head, albeit he is armed with the biggest knife around, and jas been documented to punch holes through solid metal with it.
Weakest survivor goes to Mikaela, because to me she comes across as physically weak, although her abilities with magic more than makes up for it. She's the mage of the group.
Weakest killer for me would probably be the masked kids of Legion. Maybe together they would clearly be a problem, but they're basically hyenas; not effective on their own, but as a pack both dangerous and bloody annoying.

Strongest killers are probably Myers and pyramid head since they can just ignore the entity and just kill survivors.

People who haven't played Silent Hill 2 in a while will say Pyramid Head. Realistically he's among the weakest killers. Hell, maybe even THE weakest if he's using the great knife. Weakest Surv would be Dwight I bet.
Strongest killer goes to either Nemesis, Wesker, or Sadako. Survivor would likely be Ash Williams or Chris Redfield.

About Pyramid?
He's the manifestation of James Sunderland's guilt. His helmet and knife are both meant to hinder him as well as reflect the strong emotions holding back James. I mean, just look at this first boss fight with him-
He's slow, he's struggling, and he even grunts when he does the vertical slice. Even when wielding the spear he's still slower than James and struggles to run due to the helmet. Pyramid Head is nothing like the almighty deity that the dbd fanbase has made him out to be.

People saying anything other than Myers don’t understand what Myers actually is. Canonically speaking he is Evil Incarnate. He doesn’t care about the entity, he doesn’t care about rules of the game, he just wants to kill. Nothing can stop him. There’s nothing the entity can do besides keep him there.

Nemesis is literally the most physically imposing killer in the game. Before Neme, the debate was between Oni and Clown.
Clown I think takes the cake with his high natural strength. Dark horse because I think players pass by him due to tier lists.
But that doesn’t matter because Nemesis exists.

In terms of raw strength for Killers is Nemesis, if its powerwise Sadako has an arsenal to her disposal that would put everyone to shame (15 films, 6 novels, 4 mangas, radio serials, short films etc and almost each of them introduces something to her powers).
Barring licenses then its Trapper, Oni or Clown and powerwise is Dredge.

Ehhh, I don't know about that. Ghostface was trained by a Vietnam War Vet since childhood and is supernaturally empowered by the Entity even more then usual.
Legions probably lower tier though yes.

In the entities realm the ground literally rips open with spider claws because it likes him so much. You'd have an easier time killing Trapper i'd think.

Claudette
She is in the trials of her own free will.
The Entity fears her, thus, does nothing in order to not incur her wrath.
She enjoys striking fear into the hearts of killers. Just looking at her visage fills them with anguish.
She enjoys striking fear into the hearts of survivors and sabotaging them from within. Just standing in a corner behind a stone fills survivor hearts full of despair.

New Killer: The Bush
She can teleport between map foliage as long as no one has direct line of sight on her. Survivors who witness her emerging from a bush get Spooked (healing efficiency reduced by 50%, and can only heal within 16m of the edge of the map.)
Terror radius is just a bunch of overlapping injured Claudette noises and failed skill checks.

Let's see Lore wise... (If I can remember it all)
Strong Killers:
Mickey- for sure... he's here to Kill nothing more nothing less
I could say Nurse (depending on which lore one fallows)
Oni- (one of those Killers I can't remember the lore for)
Pinhead- comes from Hell (sorry no other way to put it)
Strong Survivors:
Ash- Fighting monsters is a hard job
Freddie's Survivor (I can't spell his name)- going up against Freddie is a lot harder then one thinks (IMO)
Leon and the rest of the RE Survivors- going up against Umbrella for as long as they did is hard
Weak Killers:
Spirit aka Rin- she had to be dead for her to get her power
Hag- was kidnapped by cannibals and was almost eaten alive
Pig aka Amanda- was a junkie and became obsessed with John Kramer... oh and she had to die in order to be in the Fog
I could put Nurse here as well- depends on which lore one fallows
Legion- teenagers... not much else to say
Weak Survivors:
Dwight- Office worker who threw a prank on his boss (If I remember right)
Det. Tapp- worked for so long to catch John Kramer but died due to his obsession (only seems fair)
Claudette- Nothing in her lore proves she was strong... but knowledge could move her up
And I'm going to add an Middle section (even though it wasn't asked for)
Middle Killers:
Hillbilly- being raised the way he was... and the end result
Bubba- same as Hillbilly (the irony)
Clown- started killing animals then worked his way to humans
The rest of the cast... It's turned into a long opinionated post (sorry)
Middle Survivors:
The ones that weren't listed in strong and weak

It's blight, he has experimented on pretty much all of the other relevant killer's and Myers has never shown any remarkable feat's he just happen to has add-ons that can kill. At the very least he's stronger than any killer who has a blighted skin.
