I, uh... Well, I'm a firm believer that if they really want to, BHVR can make anything work, but uh... DDLC might be where I think they couldn't.
These two are literal just game characters Like, in the canon of their own game, they are nothing but 1's and 0's.

I wouldn't be mad if it was anime (like Sachiko from Corpse Party- yes it's also a game, I know) but, like- anime is the animation form, and DDLC is a Visual Novel. They're two completely seperate forms of media lmao.
In case people somehow haven't seen Doki Doki Literature Club and would like to, spoilers.
Doki Doki Literature Club, a visual novel/dating sim released for free on Steam in 2017 which featured one of the characters (Monika) being gifted sentience that she existed inside of a video game, and so to date the player she messes with the other character files, leading to... unfortunate accidents. Such as one of the girls, Sayori, hanging herself, or another one, Yuri, stabbing herself to death as you sit with her corpse for a weekend.
The game was released for consoles, and re-released for Steam, in 2021 as "Doki Doki Literature Club Plus" or "DDLC+", which features new bonus features like side stories of the characters (before Monika was granted sentience) artwork, soundtracks, and a lot of foreshadowing for the next project.
(A key gameplay feature of DDLC and DDLC+ is entering the game files to see what has been changed, and in DDLC+ this is taken to a new extreme).

While I don't agree with the OP and I think there's no way the next DLC is anime based, I'm quite sure we'll get it one day. Devs themselves said they could use anime as well if the idea of the killer is strong enough.

Look, I like ddlc too, with Sayori being my favorite as she has absolutely no idea whats going on half the time and I feel that cause I am stupid and never know whats going on to the point where you can consider it ‘cute’, but no
DDLC is a horror novel game with no real gameplay outside of picking options, lets just keep the doki doki where it belongs, inside of its novel
but if it is anime, don’t expect it to just be that anime, as scooby doo is considered anime due to its art style

Since this week is SawCon, the only possible anime killer could be Jigzaw. You could make a whole Saw Chapter and include Hijikata and Sogo as survivors.

Anime could work but it needs to be something really recognizable and terrifying. DDLC doesn't fit the bill since its just schoolgirls who know they're just computer code. Yet even the most popular horror anime (Death Note, Deadman Wonderland, Tokyo Ghoul, etc.) don't have an iconic pick for a Killer. I don't see it happening anytime soon.
Also, we already have 2 Japanese Killers (Spirit and Oni) and Trickster's trailer was done in a very anime-esque style.
If you look at the leaks, the next Killer is likely a solo licensed Killer. Most obvious picks are Candyman (no good choice for Survivor + movie comes out in 2-3 weeks) or Springtrap (also no iconic Survivor, recent retirement of Scott Cawthorn opens up new possibilities).
