Unable to re-enable Cross-play in Settings
I disabled cross-play preemptively without realising how much it would actually affect my queue time (it states that having it disabled may result in longer wait times, so I'm assuming switching it back on might help a little). However, when I go into my settings to re-enable it, I'm unable to. Has anyone else had this issue? I'm playing on PC through Steam, if that helps. Thank you.

Were you accessing the settings from the main menu? That's the only place you can adjust that particular setting. It won't work if you're "in a lobby".

Queue times differ depending on your rank. It suggests that they will be longer when disabled... but that isn't always the case when you're at say... Rank 20 (Grey Ranks) or Rank 16 (Yellow Ranks).
If you're at those ranks... than your queue times will usually be very long since we're VERY VERY late into rank reset. And by late, I mean the next rank reset is in 3 days...
Don't be surprised if say... you have to wait 10-15minutes per lobby at Yellow or Grey Ranks as a Survivor/Killer... this is expected at those ranks.
But!
If you feel disabling cross-play is causing this, than you can re-enable it through the settings menu (the place where you disabled it in the first place). There, you can press on the box next to it and re-enable it by checking off the box with a check mark.
Try closing the game and re-opening it (if you didn't already do this before I send this reply) and if that still doesn't work than try contacting support through the link I will be providing below.
Here's the support link: https://support.deadbydaylight.com/hc/en-us
If it still doesn't re-enable than submit a ticket with the support team so they're aware of this issue. It should re-activate when you click on the box, but if it doesn't than either what Fibijean said above is the issue OR if it still doesn't work in the main menu than do submit a ticket.

Thank you both for your answers. I was accessing the settings menu from a lobby which is why it wouldn't let me enable it again. I didn't realise that that would affect it in any way.
I also hadn't considered that my queue times might be longer due to my rank. I don't play too often these days so I'm at a pretty low rank, so that could also be factoring into the wait times. Thank you :-)

No problemo.
I too don't know why it doesn't re-enable itself UNLESS done in the main menu options tab... but regardless i'm glad your issue is resolved.
